چکیده
DNAJC25 gene is a member of the HSP40 (DNAJ) family, and it was suggested as tumor suppressor in hepatocellular carcinoma. The aim this study to analyze expression, genetic/epigenetic alterations, prognostic value breast cancer. transcript levels are upregulated BT-20 ZR-75-1 cell lines downregulated MDA-MB-231 line compared non-tumorigenic mammary epithelial (MCF 10A) (P< 0.05). According UALCAN in-silico tool, clinical cancer samples show significantly reduced mRNA relative normal (P=1.47e-02). Kaplan–Meier plotter tool shows that high expression favorable for post-progression survival (P=0.0035). Point mutations or copy number variations uncommon samples. Combined bisulfite restriction analysis (COBRA) results showed promoter not methylated lines. Promoter hypomethylation also observed (Beta-value < 0.25). In conclusion, candidate having limited biomarker potential Functional studies essential reveal its role carcinogenesis.

A candidate tumor suppressor gene in human breast cancers.
We have isolated a candidate gene (designated Brush-1) located at 13q12-q13, proximal to the retinoblastoma gene (RB1). Brush-1 codes for a 4.7-kilobase mRNA expressed at high levels in normal breast epithelium but drastically reduced in 6 of 13 breast cancer cell lines. RB1 mRNA expression is at normal levels for 5 of these 6 lines suggesting a greater importance of Brush-1 for breast cancer. ...
متن کاملDBC2, a candidate for a tumor suppressor gene involved in breast cancer.
A previously uncharacterized gene, DBC2 (deleted in breast cancer), was cloned from a homozygously deleted region at human chromosome 8p21. DBC2 contains a highly conserved RAS domain and two putative protein interacting domains. Our analyses indicate that DBC2 is the best candidate tumor suppressor gene from this region. It lies within the epicenter of the deletions and is homozygously deleted...
متن کاملDown-Regulation of ECRG4, a Candidate Tumor Suppressor Gene, in Human Breast Cancer
INTRODUCTION ECRG4/C2ORF40 is a potential tumor suppressor gene (TSG) recently identified in esophageal carcinoma. Its expression, gene copy number and prognostic value have never been explored in breast cancer. METHODS Using DNA microarray and array-based comparative genomic hybridization (aCGH), we examined ECRG4 mRNA expression and copy number alterations in 353 invasive breast cancer samp...
متن کاملANX7, a candidate tumor suppressor gene for prostate cancer.
The ANX7 gene is located on human chromosome 10q21, a site long hypothesized to harbor a tumor suppressor gene(s) (TSG) associated with prostate and other cancers. To test whether ANX7 might be a candidate TSG, we examined the ANX7-dependent suppression of human tumor cell growth, stage-specific ANX7 expression in 301 prostate specimens on a prostate tissue microarray, and loss of heterozygosit...
متن کاملA candidate tumor suppressor gene, H37, from the human lung cancer tumor suppressor locus 3p21.3.
Frequent allelic loss and homozygous deletions within chromosome 3p in human lung cancers have suggested that the 3p21.3 (370-kb) region contains a critical tumor suppressor gene(s) (TSG). With the exact identity/characteristics of such a gene(s) still unconfirmed, a lack of inactivating structural mutations in the expressed genes contained within this region may indicate that the 3p TSG(s) do ...
